Testing investigational treatment for adults with history of heart attack or heart stent and elevated lipoprotein(a)

If you have a history of heart attack or heart stent placement and elevated lipoprotein(a) levels, you may be eligible for a cardiovascular research study. This study is testing an investigational treatment for adults ages 35-80 with stable coronary artery disease and elevated Lp(a) ≥200 nmol/L.

Coronary artery disease (CAD) affects millions of adults and occurs when plaque builds up in the arteries that supply blood to the heart. This plaque buildup narrows the arteries and reduces blood flow to the heart muscle, which can lead to chest pain (angina), shortness of breath, heart attacks, and other serious complications. Many people with coronary artery disease have had a heart attack (myocardial infarction) or have received heart stents to open blocked arteries.

Lipoprotein(a), or Lp(a), is a type of cholesterol particle in your blood that is largely determined by your genes (inherited from your parents). High Lp(a) levels significantly increase your risk of

heart disease, heart attacks, and stroke, even if your regular cholesterol levels (LDL and HDL) are well-controlled with medications like statins. Unlike regular cholesterol, Lp(a) levels don’t respond well to diet, exercise, or standard cholesterol medications. Currently, there are limited treatment options specifically targeting Lp(a).

This study (OCEAN(a)-CCTA) is evaluating the safety, tolerability, and effect of an investigational treatment (olpasiran) on coronary artery plaque burden as measured by advanced heart imaging called coronary computed tomography angiography (CCTA). It will be administered as an injection once every 12 weeks (quarterly) at the study site by trained study staff for approximately 18-20 months. You’ll receive expert cardiovascular care and monitoring, including coronary CT scans at the beginning and end of the study to measure changes in plaque buildup in your coronary arteries.

Important: You must have a history of heart attack (myocardial infarction) and/or heart stent placement (percutaneous coronary intervention or PCI), and your coronary CT scan must show coronary artery disease with at least 20% narrowing in at least one major vessel. Study-related care, tests, and investigational treatment are provided at no cost. Compensation may be available.

You may qualify if you:

• Are between 35 and 80 years old
• Have a history of heart attack (myocardial infarction) and/or heart stent placement (PCI)
• Have elevated lipoprotein(a) levels (Lp(a) ≥200 nmol/L) – will be tested during screening
• Have coronary artery disease visible on CT scan (at least 20% narrowing in at least one major vessel)
• Have been on stable cholesterol-lowering medications for at least 2 weeks
• Are willing to attend study visits every 12 weeks for injections
• Are willing to have coronary CT scans at the beginning and end of the study
• Are in stable condition (no recent heart attack or stent in past 3 months, no acute coronary syndrome in past 12 months)

Adults with Heart Disease and Elevated Lp(a) Needed for Cardiovascular Research Study

Are you an adult with a history of heart disease? is conducting a clinical research study testing an investigational treatment to reduce Lp(a), a genetic cholesterol risk factor associated with increased cardiovascular risk.

About the Study:

This is a Phase 2 research study evaluating the safety and effectiveness of investigational treatment, an investigational treatment designed to reduce elevated Lipoprotein(a) [Lp(a)] levels. Lp(a) is a genetic form of cholesterol that increases the risk of heart disease, and there are currently limited treatment options available.

Who May Qualify:

Adults ages 18-80 who:

• Have a history of cardiovascular disease (heart attack, coronary stent, bypass surgery, stroke, or peripheral artery disease)
• Have elevated Lp(a) levels (≥175 nmol/L)
• Have BMI between 18.5 and 40
• Are on stable heart medications (statins, etc.) for at least 4 weeks
• Are willing to attend study visits over approximately 2 years

What to Expect:

• Receive investigational treatment (investigational treatment) or placebo via subcutaneous injection
• Three different dosing options: every 26 weeks or every 52 weeks depending on study group
• Regular study visits for monitoring and assessments
• Blood tests to measure Lp(a) levels and safety monitoring
• Extended follow-up period to assess long-term effects
• All participants eventually receive active treatment (cross-administration design)

Study Duration:

Approximately 2 years (104 weeks) total:

• Prescreening to check Lp(a) levels
• Screening period (up to 21 days)
• Treatment period (52 weeks) with regular visits
• Extended follow-up (up to 52 additional weeks)

Compensation:

Participants will receive compensation for their time and participation. All study-related care, procedures, and investigational treatment are provided at no cost.
